Measurement of spreading depolarization in Drosophila melanogaster. After immobilizing the fly, a reference electrode is inserted into the abdomen (bottom) and two glass microelectrodes (from left and right) are inserted into its brain. With one electrode we monitored the electrical activity of the brain during cooling and with the other the K+ concentration in the extracellular fluid in the brain. Spreading depolarization is characterized by an abrupt drop in electrical activity and a surge in extracellular K+ concentration and this occurs at the temperature where these insects go into coma.
